Target Validation in Synthetic Lethality Overview

Take part in early exploration and discovery of future targets! This internship is aimed at biologically validating targets that are nominated by in-house functional genomics screens. The intern will work closely with scientists in Discovery Oncology, Biology to test the effect of CRISPR/Cas12 knockouts of gene targets that are potentially druggable and clinically relevant. The intern will thus become familiar with state-of-the-art technologies and techniques of gene knockouts through electroporation or lentiviral vectors, protein detection and cell viability readouts that are integral for target validation which is the starting point for drug discovery pipeline projects.
This internship is located in South San Francisco, CA.
Key responsibilities include:
- Cell culture of human cell lines
- Lentivirus generation (BSL2) (optional)
- Lentiviral transduction and selection of cell lines (optional)
- Electroporation of CRISPR/Cas12 crRNAs
- Flow cytometry
- Protein purification and detection (Simple Western)
- Cell viability assessment (Cell Titer Glo (CTG))
- Cell proliferation and tracking (Incucyte)
- DNA/RNA purification
- Compiling and presenting data to manager
Qualifications
Minimum Qualifications
- Currently enrolled in university, pursuing a Master’s or PhD in Biology, Cell Biology, Biomedicine or other related field
- Returning to university for at least one semester following the internshipÂ
- Cell culture experience (aseptic technique)
Preferred Qualifications
- Exposure to CRISPR/Cas9 or CRISPR/Cas12 technology
- Biosafety level 2 (BSL2) qualification is a plus
- Electroporation (Invitrogen Neon or Lonza 4D Nucleofector)
- Experience with plate based (96wp, 384wp) assay formats
- Flow cytometry (BD X-20 Fortessa, Canto or similar)
