The focus of our lab is on cancer drug discovery using fragment-based methods and structure-based design. To accomplish this goal, we have assembled a multidisciplinary team that includes structural biologists, medicinal chemists, and cell biologists. In our lab, we clone, express, and purify proteins, conduct fragment-based screens, determine the three-dimensional structures of protein/ligand complexes using NMR and/or Xray crystallography, design and synthesize compounds, and test compounds in primary and secondary biological assays. The cancer drug targets that we are pursuing are highly validated but technically challenging such as KRas, Mcl1, WDR5, HIF1a, and proteins in the Wnt pathway.
Duties and Responsibilities
- Assist in Research and Development
- Under moderate supervision, prepare bacterial cell culture in flasks and fermenter using both rich and minimal media for normal and isotopically labelled protein expression.
- Conduct expression optimization, solubility assay, cell homogenization and protein purification
- Vanderbilt University Biochemistry Job Design of protein constructs, cloning, site-directed mutagenesis, plasmid purification, sequencing and biophysical characterization of purified proteins
- Preparation and maintenance of chromatographic columns and purification systems, troubleshooting and placing a service request
Contribute to the effective operation of the lab by sharing the responsibilities of - preparing buffers and reagents, ordering supplies, and maintaining equipment
Remain current on literature, ideas, and relevant laboratory techniques in oncology and drug discovery - Perform routine and experiment-based maintenance, protein expression, and purification
- Duties include media preparation, maintenance of competent cells, chromatographic columns, and purification instruments
- Run SDS polyacrylamide gel electrophoresis to assess protein purity of elution fractions
- Techniques include the transformation of expression plasmids and protein functional assay
- Perform additional lab duties to ensure the laboratory is prepared for experiments
- Ensure the most effective operation of the laboratory

- Maintain equipment
- Work with lab mates to maintain sterile & organized environment in cell culture facility
Organize, document, and maintain freezers, refrigerators, cell line storage
